Abstract

Research on antioxidant activity test of Kadamba leaf extract and fraction (Mitragyna speciosa KORTH.) using ABTS scavenging method has been conducted. This study aims to determine the secondary metabolites of antioxidant activity of Kadamba leaf extract and fraction against ABTS radicals. Antioxidant activity test was conducted using ABTS scavenging method. Kadamba leaves were extracted using methanol solvent and fractionated using n-hexane, ethyl acetate, and n-butanol solvents. The results showed that the IC50 values ​​of crude extract, n-hexane fraction, ethyl acetate fraction, and n-butanol fraction were 81.417 ppm; 60.050 ppm; 96.059 ppm; and 160.673 ppm, respectively. Methanol extract, n-hexane fraction, and ethyl acetate fraction have strong antioxidant activity, namely 50 ppm > IC50 < 100 ppm, while the n-butanol fraction has weak antioxidant activity, namely IC50 > 150 ppm.
