Abstract

Mangostin merupakan senyawa aktif utama dari kulit manggis (Garcinia mangostana L.) yang memiliki potensi farmakologis luas. Namun, pemanfaatannya masih dibatasi oleh kelarutan air yang rendah dan bioavailabilitas yang buruk. Pengembangan sistem penghantaran berbasis nanoemulsi menjadi salah satu strategi untuk mengatasi kendala tersebut. Kompleksitas formulasi menuntut pendekatan optimasi yang efisien, sehingga Design of Experiments (DoE) lebih unggul dibandingkan metode trial-and-error konvensional. Penelitian ini bertujuan meninjau secara sistematis penerapan strategi DoE dalam optimasi formulasi penghantaran mangostin, khususnya nanoemulsi, SNEDDS, dan nanoemulgel, untuk memperoleh karakteristik fisikokimia dan stabilitas yang optimal. Penelusuran literatur dilakukan pada database terakreditasi dengan fokus pada studi yang menggunakan rancangan DoE. Hasil tinjauan menunjukkan bahwa Simplex Lattice Design dan Box-Behnken Design merupakan metode yang paling banyak digunakan. Formula optimum umumnya memanfaatkan Virgin Coconut Oil atau minyak kelapa sawit sebagai fase minyak, serta kombinasi Tween 80 dengan PEG 400 atau Span 80 sebagai surfaktan dan ko-surfaktan. Faktor kritis yang paling memengaruhi respons meliputi konsentrasi minyak, rasio surfaktan campuran, dan waktu sonikasi. Optimasi berbasis DoE terbukti menghasilkan sistem dengan ukuran droplet nanometrik, PDI rendah, efisiensi penjeratan tinggi, dan stabilitas fisik yang baik.
