Peningkatan Laju Disolusi Ketoprofen yang Diformulasikan dengan Metode Self Emulsifying Drug Delivery System (SEDDS)

Abstract

Setelah pemberian obat secara oral, disolusi obat merupakan syarat absorbsi suatu obat ke sirkulasi sistemik. Berdasarkan klasifikasi Biopharmaceutical Classification System(BCS), ketoprofen termasuk dalam kelas II yakni tingkat kelarutan yang rendah dengan karakteristik permeabilitas yang baik (permeabilitas yang efektif pada manusia untuk ketoprofen 8.70×10-4cm/s). Self-Emulsifying Drug Delivery System (SEDDS) dapat meningkatkan bioavailabilitas obat yang memiliki kelarutan yang rendah tetapi permeabilitas yang tinggi. Oleh karena itu, untuk meningkatkan laju disolusi maka dilakukan formulasi ketoprofen dengan metode SEDDS. Dilakukan uji kelarutan ketoprofen pada beberapa jenis minyak dan surfaktan yaitu minyak jagung, minyak biji bunga matahari, minyak kedelai, asam oleat, tween 80 dan span 80 dengan metode saturation solubility. Dibuat tiga formula yang mengandung zat aktif ketoprofen 50 mg, tween 80:span 80 (3:1) sebagai surfaktan dengan konsentrasi 30% (F1), 45% (F2), 60% (F3) dan minyak sebagai pembawa. Hasil uji disolusi menunjukkan bahwa ketoprofen yang diformulasikan dengan metode SEDDSdapat meningkatkan laju disolusi ketoprofen, persentase terdisolusi tiap formula setelah 1 jam adalah 62,91% (F1), 78,09% (F2), 100,6% (F3), dan 49,95% pembanding. Analisis statistika menggunakan rancangan acak lengkapmenunjukkan perbedaan nyata antara laju disolusi (F1), (F2) dan (F3) dengan pembanding. Formula ketoprofen dengankonsentrasi surfaktan 60% (F3) menunjukkan laju disolusi tertinggi dibandingkan dengan (F1) dan (F2) serta dengan pembanding.
